DNA microarray diagnostics

DNA microarrays consist of DNA sequences (probes) applied to a solid carrier such as glass in microscopically small spots at defined positions. When a patient's DNA contains segments that match the probes, the complementary regions hybridize. This binding is read by computer and evaluated as a positive signal.

Features

1 · Sample preparation

  • The patient's DNA is extracted from blood using DNA isolation kits

2 · Amplification (PCR)

  • The target DNA sections are amplified million-fold by polymerase chain reaction
  • Two primers define the region to be copied; if the target sequence is present it is copied exponentially
  • PCR products are labelled with a fluorescent dye for detection on the microarray

3 · Microarray hybridization

  • PCR products are mixed with hybridization buffer and incubated with the microarray
  • Binding to complementary probes is measured via the fluorescence signals of the spots
